Among these proteins, Akap4, Atp5a1 and Eno1 were downregulated in the stressed group compared with the control group, whereas Pkm2, Prss2, Ywhaz, Acsm2 and Myl9 were upregulated. To assess the effects of chronic stress on the expression of proteins in the testis, we constructed 2‐DE maps with high resolution for the testis at day 21, which showed that 375.5 ± 15.5 and 407.5 ± 17.5 protein spots (mean ± SD) differentiated the control and stressed groups (Figure 3A). PDQuest professional software was used for qualitative and quantitative mapping analysis for the differential spots from testis tissue proteins in the stressed group and control group. The cell viability was estimated with an FLUO star Omega plate reader (BMG LABTECH GmbH) at 492 nm. Antibodies were incubated at 4°C overnight after antigen retrieval, and the secondary antibody was incubated the next day. Paraffin sections (5 µm) of the testis were deparaffinized and rehydrated with xylene and gradient ethanol (100%, 95%, 90% and 80%), and endogenous peroxidase activity was eliminated. Pompe et al. conducted a study on men who underwent EBRT for 2 years and revealed that 75% of patients underwent a significant decline in testosterone levels with up to 40% increase in the rates of biochemical hypogonadism. Exposure to RF EMFs originating from gadgets like microwave ovens and cell phones could generate seminal ROS leading to oxidative stress 134,152. The fluctuations were detected due to overproduction in radiation-induced ROS levels, which clearly indicates infertility in male rats after the exposure . Moreover, DEHP dramatically changed the manner of interaction between LCs and other cells, and the most significant type was the cell-cell contact, which included NECTIN, APP, CADM, and CD39. Leydig cells (LCs) injury is often irreversible upon discovery; hence, early identification of risk factors for injury is crucial. These disturbances may lead to impaired sexual potency, eventually causing disruption of psychological health in affected males. According to the guidelines of the International Consultation for Sexual Medicine (ICSM), TRT is advisable when the level of total testosterone is 281 and/or in the presence of LOH symptoms, with these criteria being consistent across various countries.|Together they address two fundamental aspects of healthy aging from different angles. Align directly targets gut health for digestive comfort and nutrient absorption. The single daily capsule fits easily into busy routines while supporting both digestive comfort and overall health through the gut-immune connection. For high-stress professionals, it buffers stress-induced digestive upset. Digestive issues often emerge or worsen in your 40s due to stress, dietary changes, and natural microbiome shifts.|Specifically, antioxidant components were scored as high (2), medium (1), and low (0) levels, respectively, while pro-oxidant components were scored inversely. In this study, each component of the Oxidative Balance Score (OBS) was assigned a score based on its tertile distribution. Physical activity was defined using the formula from this literature (18). Second, lifestyle OBS includes alcohol consumption, serum cotinine, BMI, and physical activity.|The subsequent decline in testosterone levels may disrupt NOS activity in the penile tissues, further suppressing NO production, ultimately giving rise to ED . During the condition of oxidative stress, ROS tends to stimulate producing inhibin, along with testicular estradiol (E2), which results in reduced testosterone synthesis . Through the crosstalk between the HPG and HPA axes, cortisol negatively affects the LH secretion from the anterior pituitary, which is followed by decreased testosterone production by the Leydig cells. However, ROS interfere with the communication between the Leydig cells and the HPG axis, lowering testosterone levels . During oxidative stress, biomolecules undergo indiscriminate damage, leading to loss of function and even cell death. Fat accumulation due to obesity leads to increased oxidative stress levels in males (38).|Kesari et al. also suggested an adverse impact of cell phone radiation on semen quality, particularly a decline in sperm count, motility, viability, morphology and increase in apoptosis . Hence, the decline in testosterone level could possibly hamper the process of spermatogenesis 148,149. While exerting such effects, radiation may first disturb the Ca2+/CaMKI turnover in Leydig cells and thereafter inactivate the RORα clock gene.|Increased production of LTH resulting from excessive ROS generation may also create hindrance in the HPG axis. Testosterone binds to ABP and brings about the functional response required for spermatogenesis. Inhibin is a negative regulator of FSH secretion, and excessive release of this hormone inhibits the normal spermatogenic process. It is also expressed in the testicular tissues, which regulates local processes that are strategically regulated by pituitary GH.|BPA-mediated ROS generation may also suppress testosterone activity and spermatogenic processes directly. As a result, the plasma concentration of LH fails to rise in response to the decreased testosterone levels, implying that PCB also disrupts a pituitary feedback mechanism . First, steroidogenic enzyme activity within Leydig cells is reduced, ultimately leading to decreased biosynthesis of androgens. Based on testosterone levels and E2, a correlation of PFOS and perfluorooctanoic acid (PFOA) levels with the delay of puberty in children (boys and girls) has also been demonstrated . Detrimental effects of DEHP on Leydig cell steroidogenesis occurs through the modulation of testosterone-biosynthetic enzyme activity. Devitalization of pituitary LH secretion during development adversely affects the acquisition of steroidogenic capacity, thereby decreasing testosterone biosynthesis by Leydig cells .|Many lifestyle habits can also significantly influence testosterone levels. Basic research has also demonstrated that the combined application of zinc and selenium enhances testosterone levels by strengthening antioxidant mechanisms (30). However, studies have also revealed that men adhering to a low-fat diet tend to exhibit lower testosterone levels (28). The Mediterranean diet, which includes a variety of antioxidant-rich components, has been shown to significantly elevate testosterone levels when adhered to over the long term (26). As shown in Figure 3, no significant association was observed between OBS and testosterone deficiency across the stratified age groups. This study further stratified OBS into lifestyle OBS and dietary OBS to separately examine their associations with low testosterone in males. Total testosterone levels in serum were measured using isotope dilution liquid chromatography tandem mass spectrometry (ID-LC–MS/MS).} Chlorpyrifos also reduces the concentration of sialic acid, which then inhibits Leydig cells. Consequently, the entire HPG axis is disrupted, leading to reduced testosterone production. Lowering testosterone concentration, in turn, impacts the negative feedback regulation of FSH, further hindering the process of spermatogenesis . It probably converts itself into chlorpyrifos oxon and decreases the level of serum testosterone and sperm count . An increase in testicular cholesterol can lower the concentration of androgens and consequently hamper spermatogenesis . This is also in agreement with recent studies (Buchanan et al. 2001; Duckworth et al. 2001; see also Mougeot et al. 2004), which have shown that the cost of high testosterone concentration is not exclusively based on immunosuppression, as proposed by the immunocompetence handicap hypothesis (Folstad & Karter 1992). Androgens increase angiotensin II production, which regulates vasoconstriction by producing peroxides from NADH- and NADPH-oxidase (Reckelhoff 2001). Alternatively or additionally, the effect could be mediated by the role of androgens in the regulation of blood pressure. To assess the sensitivity of the results to these extreme values, we also ran the analyses excluding birds that had testosterone values higher than 20 ng ml−1. In this study, the highest value for control birds (with empty implants) was 7.66 ng ml−1, whereas the highest value for T-individuals was 35 ng ml−1. Exposure to PM2.5 has also been linked to inflammation and oxidative stress and hence may potentially cause ED of vascular origin through veno-occlusive or arteriogenic pathways . ROS is an important factor that induces cell death and spermatogenic cell damage 223,224. Elevated exposure to PM2.5 increases ROS production in the body, which may enhance TNF-α level. Psychological, physiological, hormonal, pathological, environmental, and nutritional stressors are believed to play a major role in the pathogenesis of ED . Another common male sexual disorder is ED that involves an alteration in any of the components of erectile response–organic, relational, and psychological . Some epidemiological studies showed that community-dwelling men above the age of 30 years are characterized by an annual decrease of 0.5–15% in the concentration of circulating testosterone and a 2–3% decrease in the concentration of free testosterone . It not only hampers the process of spermatogenesis but also disturbs normal reproductive physiology.
